目的:提高抗肿瘤抗生素在发酵液中的含量,并进行分离纯化。方法:采用微生物法、DNA断裂法分析抗肿瘤抗生素在发酵液中的含量,以响应曲面法优化发酵培养基。结果:通过改变发酵培养基的组份及优化其配比,新型产生菌发酵液中抗肿瘤抗生素的含量提高约30倍。在其后的实验中运用优化的培养基发酵,从中分离纯化出高效抗肿瘤抗生素,经初步测定相对分子质量为1367。结论:培养基组份及其配比影响其代谢产物的组成及含量,响应曲面法可以用相对较少的实验获得优化的发酵条件,提高抗肿瘤抗生素的含量。
Objective: To improve the content of antitumor antibiotics in fermentation broth and to separate and purify. Methods: The contents of anti-tumor antibiotics in the fermentation broth were analyzed by the method of microorganism and DNA fragmentation, and the fermentation medium was optimized by the response surface methodology. Results: By changing the components of fermentation medium and optimizing the ratio, the content of anti-tumor antibiotics in the new fermentation broth increased about 30 times. In the subsequent experiments using the optimized fermentation medium, from which highly efficient anti-tumor antibiotics were isolated and purified, the initial determination of the relative molecular mass of 1367. Conclusion: The composition of media and its composition affect the composition and content of its metabolites. Response surface methodology can be used to obtain optimized fermentation conditions and improve antitumor antibiotics with relatively few experiments.
