目的:建立一测多评法(QAMS)同步测定菊花中绿原酸、木犀草苷、槲皮素和金合欢素4种成分的含量,并验证该方法在菊花质量评价中的应用价值。方法:采用Shim-pack C18(250 mm×4.6 mm,5μm)色谱柱;流动相为乙腈(A)-0.1%磷酸溶液(B),梯度洗脱;体积流量1.0 m L/min;柱温30℃;进样量10μL;检测波长348 nm。以绿原酸为内参物,建立其与木犀草苷、槲皮素和金合欢素的相对校正因子,并计算各成分含量,实现一测多评。同时采用外标法验证一测多评法的准确性和可行性。结果:绿原酸与木犀草苷、槲皮素和金合欢素的相对校正因子分别为0.6865、0.9976、0.6665,RSD(n=6)分别为0.1439%、0.2512%、0.3971%;一测多评法与外标法含量测定结果无显著性差异。结论:以绿原酸为内标同时测定木犀草苷、槲皮素和金合欢素的一测多评法可用于菊花的定量分析。
OBJECTIVE: To establish a multi-assessment method (QAMS) for the simultaneous determination of four components in chrysanthemum, including chlorogenic acid, luteolin, quercetin and acacsin, and verify the application value of the method in quality evaluation of chrysanthemum. Methods: A Shim-pack C18 (250 mm × 4.6 mm, 5 μm) column was used. The mobile phase consisted of acetonitrile-A 0.1% phosphoric acid solution (B) ℃; injection volume 10μL; detection wavelength 348 nm. Chlorogenic acid was used as an internal control to establish its relative correction factor with luteolin, quercetin and acacetin, and to calculate the content of each component to achieve a test for multiple assessment. At the same time, the accuracy and feasibility of using multiple external evaluation methods to validate a multi-evaluation method are verified. Results: The relative correction factors of chlorogenic acid, luteolin, quercetin and acacetin were 0.6865, 0.9976 and 0.6665 respectively, and the RSD (n = 6) were 0.1439%, 0.2512% and 0.3971% There was no significant difference between the method and the external standard method. Conclusion: The simultaneous determination of luteolin, quercetin and acacetin by chlorogenic acid can be used for the quantitative analysis of chrysanthemum.
