高效快速的品种(系)DNA指纹图谱鉴定,可为辅助牛鞭草属品种(系)DUS测试和知识产权保护等提供科学依据。本研究从54对SSR引物中筛选出20对引物对牛鞭草属25个DUS测试材料进行鉴定分析。结果表明,20对SSR引物对牛鞭草属25个DUS测试材料共扩增出253条清晰条带,其中多态性条带241条,多态性条带比率(PPB)为95.26%。每对引物的多态信息含量(PIC)为0.599~0.900,平均为0.760。UPGMA聚类分析表明,牛鞭草属25个材料在相似系数为0.668处可分为三大类群。20对SSR引物能鉴别10~22个牛鞭草属材料,引物DHSSR7、引物DHSSR11和引物DHSSR15的有效组合可鉴别所有供试材料。以3对SSR引物扩增的电泳图谱为基础,建立了牛鞭草属25个DUS测试材料的指纹图谱标准模式图,能够快速区分各个供试材料。
Efficient and rapid identification of varieties (lines) DNA fingerprinting can provide a scientific basis for assisting the Verbena genus DUS testing and intellectual property protection. In this study, 20 pairs of primers were selected from 54 pairs of SSR primers to identify 25 DUS test materials of the genus Verbena. The results showed that a total of 253 clear bands were amplified from 25 DUS test materials of Verbenaceae by 20 pairs of SSR primers, of which 241 were polymorphic and 95.26% were polymorphic bands (PPB). The polymorphic information content (PIC) of each pair of primers ranged from 0.599 to 0.900 with an average of 0.760. UPGMA cluster analysis showed that 25 species of the genus Verbena could be divided into three groups at a similarity coefficient of 0.668. Twenty pairs of SSR primers were able to identify 10 to 22 Verbena materials. An effective combination of primer DHSSR7, primer DHSSR11 and primer DHSSR15 identified all the tested materials. Based on the electropherogram of three pairs of SSR primers, the standard model of fingerprinting of 25 DUS test materials of the genus Verbena was established, which can quickly distinguish the tested materials.
