目的:探讨降糖防聋方对体外高糖培养成骨细胞骨钙素的影响。方法:采用改良的组织块法分离培养新生大鼠颅骨成骨细胞,用不同浓度的降糖防聋方含药血清及不含降糖防聋方含药血清的α-MEM培养液加入到成骨细胞高糖(糖终浓度为300 kg·L-1)培养体系,作用不同时间,应用放免法测定细胞骨钙素(BGP)的水平。结果:100,50,10 mg·L-1的降糖防聋方含药血清对体外高糖培养成骨细胞具有明显的促进作用(P<0.05),在较高浓度时对成骨细胞增殖无明显抑制效应。结论:降糖防聋方含药血清对高糖培养成骨细胞骨钙素有不同程度调节改善作用。
Objective: To investigate the effects of Jiangtanggandefang on osteocalcin in cultured osteoblasts in vitro. Methods: Newborn rat calvarial osteoblasts were isolated and cultured by modified tissue block method. The cells were treated with α-MEM containing different concentrations of serum containing hypoglycemic anti-deaf and serum containing hypoglycemic anti-deaf side serum Osteoblast cells were cultured with high glucose (final concentration of 300 kg · L -1) for different time. The level of osteocalcin (BGP) was measured by radioimmunoassay. Results: The serum containing 100, 50, 10 mg · L-1 hypoglycemic and anti-deaf herbs significantly promoted the osteoblasts cultured in vitro with high glucose (P <0.05) and proliferated at higher concentrations No significant inhibitory effect. CONCLUSION: The serum containing Jiangtang Fangtong Decoction has good effects on the osteoblast osteocalcin in high glucose.
