目的研究KLF14基因rs972283多态性与宁夏2型糖尿病的关系。方法用聚合酶链反应-限制性片段长度多态性的方法(RFLP-PCR),对宁夏2型糖尿病患者(DM组)和正常对照组(NC组)KLF14基因rs972283位点进行基因多态性和等位基因频率分析,测定相应生化指标。结果 KLF14基因rs972283位点多态性在DM组和NC组的分布差异无统计学意义(P>0.05);DM组内携G等位基因患者的体质指数(BMI)、腰臀比(WHR)、血胆固醇(TC)、胰岛素抵抗指数(HOMA-IR)均高于携A等位基因者,DM组内携G等位基因者胰岛β细胞功能(HOME-β)较携A等位基因者低,差异均有统计学意义(P<0.05)。结论 KLF14基因rs972283位点多态性可能与宁夏2型糖尿病发病无关,与肥胖、血脂代谢等有关。
Objective To study the relationship between rs972283 polymorphism of KLF14 gene and type 2 diabetes mellitus in Ningxia. Methods The gene polymorphism of rs972283 in KLF14 gene of type 2 diabetic patients (DM group) and normal control group (NC group) of Ningxia was studied by polymerase chain reaction-restriction fragment length polymorphism (RFLP-PCR) And allele frequency analysis, determination of the corresponding biochemical indicators. Results There was no significant difference in the distribution of rs972283 polymorphism in KLF14 gene between DM group and NC group (P> 0.05). The BMI, WHR, TC and HOMA-IR were higher than those with A allele, but those with G allele in DM group were significantly higher than those with A-allele Low, the difference was statistically significant (P <0.05). Conclusion Polymorphism of rs972283 in KLF14 gene may be related to the incidence of type 2 diabetes in Ningxia and to obesity and lipid metabolism.
