目的明确雌激素受体(ER)阴性、阳性MCF-7细胞增殖速度差异以及不同MCF-7细胞对鸡血藤黄酮类提取物SSCE抗肿瘤作用的敏感性。方法免疫细胞化学(ICC)方法确定3种MCF-7细胞(MCF-7-a、MCF-7-b、MCF-7-luc)ER表达情况,MTT法检测3种细胞的增殖速度以及对SSCE抑制肿瘤细胞生长作用的敏感性。结果 ER(+)的MCF-7-a、ER(-)的MCF-7-b和MCF-7-luc倍增时间分别为(69.95±9.37)h、(47.17±5.27)h、(44.96±10.33)h,24 h、48 h、72 h各个时间点MCF-7-luc对SSCE的半数抑制浓度均低于另外两株细胞。结论 ER(-)的乳腺癌细胞恶性增殖速度高于ER(+)的乳腺癌细胞;荧光素酶基因标记的ER(-)MCF-7细胞对SSCE抗肿瘤作用更加敏感;提示ER的表达情况与乳腺癌的恶性增殖程度有关,基因重组可能影响了细胞膜表面蛋白的表达,从而增强了细胞对药物的敏感性。
Objective To determine the difference of the proliferation rate of estrogen receptor (ER) negative and positive MCF-7 cells and the sensitivity of different MCF-7 cells to anti-tumor effects of spionensin SSCE. Methods The expression of ER in MCF-7 cells (MCF-7-a, MCF-7-b and MCF-7-luc) was determined by the method of immunocytochemistry (ICC). The proliferation of three kinds of cells was detected by MTT assay, Sensitivity to inhibit the growth of tumor cells. Results The doubling time of MCF-7-b and MCF-7-luc in ER (+) MCF-7-a and MCF-7-luc cells were 69.95 ± 9.37 h, 47.17 ± 5.27 h and 44.96 ± 10.33 h ), the half inhibitory concentration of MCF-7-luc on SSCE at 24 h, 48 h and 72 h was lower than that of the other two cells. CONCLUSION: ER (-) breast cancer cells proliferate faster than ER (+) breast cancer cells. Luciferase-labeled ER (-) MCF-7 cells are more sensitive to antitumor activity of SSCE. With the degree of malignant proliferation of breast cancer, gene recombination may affect the expression of cell surface protein, thereby enhancing the cell sensitivity to drugs.
