目的:研究肾阳虚大鼠制首乌含药血清对L02肝细胞增殖和凋亡的影响。方法:采用皮下注射氢化考的松,建立大鼠肾阳虚模型,分为生理盐水组,肾阳虚低、中、高浓度组(10.8、21.6、43.2g/kg)。连续给药14d后处死大鼠,制备含药血清,分别采用MTT法测定L02肝细胞线粒体功能、流式细胞术及荧光染色检测肝细胞凋亡状态。结果:15%肾阳虚各浓度组含药血清作用24h和48h后对L02肝细胞增殖均具有明显抑制作用,其中,肾阳虚高浓度组含药血清作用48h后细胞抑制率达24.92%(P<0.01);肾阳虚中、高浓度组的细胞凋亡率显著性升高,分别为40.90%、46.83%(P<0.01);Annexin V-FITC/PI荧光染色显示,肾阳虚中、高浓度组出现早期凋亡及晚期凋亡的细胞较多。结论:肾阳虚证为制首乌致肝损伤的靶向毒性证候,为临床上正确使用制首乌,避免不良反应的出现提供依据。
Objective: To study the effect of Radix polygoni multiflori containing Radix Astragali on the proliferation and apoptosis of L02 hepatocytes. Methods: Subcutaneous injection of hydrocortisone to establish rat kidney yang deficiency model, divided into saline group, low yang, middle and high concentration group (10.8,21.6,43.2 g / kg). The rats were sacrificed 14 days after administration, and the serum was prepared. The mitochondrial function of L02 hepatocytes was determined by MTT assay, and the apoptosis of hepatocytes was detected by flow cytometry and fluorescent staining. Results: After treated with serum of 15% Shen Yang deficiency group for 24 h and 48 h, the proliferation of L02 hepatocytes was significantly inhibited. Among them, the cell inhibitory rate of 24 h was 48.9% P <0.01). The apoptotic rates in middle and high concentration of Shenyang were significantly increased (40.90% and 46.83%, respectively) (P <0.01). Annexin V-FITC / , High concentration group appeared early apoptosis and late apoptosis more cells. Conclusion: Kidney-Yang Deficiency Syndrome is the target toxic syndrome caused by Radix Polygoni Multiflori. It is the basis for clinical use of Radix Polygoni Multiflori and the avoidance of adverse reactions.
