目的:利用计算机图像处理技术辅助建立一种新的免疫酶组织化学多重染色技术。方法:采用改良的SP法,在同一张切片上进行肝癌组织中3种已知阳性表达的蛋白(乙肝表面抗原HBs、人去唾液酸糖蛋白受体ASGPR和增殖细胞核抗原PCNA)的免疫酶组织化学多重染色,并借助计算机图像处理技术将3张图合成1张能显示多种蛋白表达情况的图。结果:3种蛋白在利用了新的免疫组化多重染色技术进行染色后,阳性信号强烈,效果明显,且不发生抗体的交叉反应,利用计算机图像处理技术处理后的图片也能客观的展示出原来图片中抗原的阳性信号的位置和表达情况,并在同一张图片上展示出3种不同蛋白的表达情况。结论:利用计算机图像处理技术及新的免疫组化多重染色技术能实现在同一张切片上标记多种蛋白的目的。
Objective: To establish a new immunoenzymatic histochemical staining technique using computer image processing technology. Methods: Immunohistochemical methods were used to detect the expression of three known proteins (HBsAg, ASGPR and PCNA) in hepatocellular carcinoma using the modified SP method. Chemical multi-staining, and the use of computer image processing technology will be three pictures into a map can show a variety of protein expression. Results: The three kinds of proteins were stained with a new immunohistochemical multi-staining technique. The positive signals were strong and the effect was obvious. The cross-reaction of antibodies was not occurred. The pictures processed by computer image processing technology could also be objectively displayed The original image of antigen positive signal location and expression, and in the same picture shows the expression of three different proteins. Conclusion: The use of computer image processing technology and a new immunohistochemical staining technique can achieve the purpose of labeling a variety of proteins on the same slice.
