用卵巢癌细胞系A2780在体外分别与化疗药物长春新碱(VCR)、顺铂(DDP)和阿霉素(ADM)作用,获得A2780/VCR、A2780/DDP和A2780/ADM3个耐药细胞亚株,通过体外细胞毒性实验(MTT法),观察A2780与药物作用后的耐受情况及耐药细胞株对10种化疗药物的敏感性。结果:三株细胞对VCR、DDP和ADM均有耐药,对10种作用机制不同的化疗药的耐药指数相差很大。提示:三株卵巢癌细胞均有多药抗药性(MDR),药物引起的耐受可分为两类型:单向性交叉耐药和双向性交叉耐药。
A2780 / VCR, A2780 / DDP and A2780 / ADM three drug-resistant cells were obtained by using ovarian cancer cell line A2780 in combination with chemotherapeutic drugs vincristine (VCR), cisplatin (DDP) and adriamycin (ADM) Strain, through the cytotoxicity test (MTT method) to observe the A2780 and drug resistance after tolerance and drug-resistant cell lines on the sensitivity of 10 kinds of chemotherapy drugs. Results: The three strains of cells were resistant to VCR, DDP and ADM, and the drug resistance index of 10 chemotherapeutic agents with different mechanism of action varied greatly. Tip: Three ovarian cancer cells have multidrug resistance (MDR), drug-induced tolerance can be divided into two types: one-way cross-resistance and two-way cross-resistance.
