目的:建立高效液相法同时测定淫羊藿饮片中朝藿定A、朝藿定B、朝藿定C、淫羊藿苷和宝藿苷I的含量,比较37批次淫羊藿饮片中5种黄酮类成分含量差异。方法:应用高效液相法进行测定:色谱柱为Luna-C18(4.6mm×250mm,5μm);乙腈-水梯度洗脱,0~29min,乙腈25%;29~30min,乙腈25%~41%;30~55min,乙腈41%;55~56min:乙腈41%~25%;56~64min,乙腈25%;流速为1mL·min-1;检测波长为270nm;进样量为10μL;柱温为30℃。结果:37批次淫羊藿饮片中朝藿定A含量在1.80~3.88mg·g-1,朝藿定B含量在3.48~20.25mg·g-1,朝藿定C含量在1.37~29.62mg·g-1,淫羊藿苷含量在3.45~8.12mg·g-1,宝藿苷I含量在1.52~5.23mg·g-1。其中以柔毛淫羊藿中朝藿定C含量最高,淫羊藿中朝藿定C、朝藿定B含量最高。结论:本实验中所建立的色谱方法可以简便、准确测定淫羊藿饮片中朝藿定A、朝藿定B、朝藿定C、淫羊藿苷和宝藿苷I的含量,重复性好;建议在建立淫羊藿饮片的质量标准时,应建立多种黄酮类成分指标评价体系,以全面控制淫羊藿饮片的质量。
OBJECTIVE: To establish a HPLC method for simultaneous determination of the content of Epimedium Astragalus membranaceus A, Zhichading B, Chaozhiding C, icariin and Abelylose I in Epimedium Herbal Pieces, and to compare 37 batches of Herba Epimedii Tablets. The difference in the content of five flavonoids. Methods: The HPLC method was used for the determination: the chromatographic column was Luna-C18 (4.6mm×250mm, 5μm); acetonitrile-water gradient elution, 0~29min, acetonitrile 25%; 29~30min, acetonitrile 25%~41% 30~55min, acetonitrile 41%; 55~56min: acetonitrile 41%~25%; 56~64min, acetonitrile 25%; flow rate is 1mL·min-1; detection wavelength is 270nm; injection volume is 10μL; column temperature is 30 °C. RESULTS: The contents of 37 batches of Epimedium decoction pieces were between 1.80~3.88mg·g-1, the content of Chaojuding B was 3.48~20.25mg·g-1, and the content of Cyndicarb was 1.37~29.62mg. · g-1, icariin content 3.45 ~ 8.12mg · g-1, Bastatin I content in 1.52 ~ 5.23mg · g-1. Among them, C content was highest in Epimedium chinensis, Sino-Kazakh dynasty, and Epimedium in middle and North Korea were the highest. Conclusion: The chromatographic method established in this experiment can easily and accurately determine the content of the two components in the epimedium-decoction decoction pieces: Chao-Ding-Ding, Chao-Ding-Ding B, Chao-Ding-Ding C, Icariin, and Benzaside I. The repeatability is good. It is recommended that when establishing the quality standards of Epimedium Herbal Pieces, a variety of flavonoid composition index evaluation systems should be established to fully control the quality of Epimedium Herbal Pieces.
