目的:建立固本明目颗粒挥发油中丁香酚含量的HPLC测定方法。方法:采用高效液相色谱法(HPLC)测定丁香酚含量,色谱柱为Waters C_(18)(250mm×4.6mm,5μm),流动相为甲醇-水(55∶45,V/V),流速为1.0mL/min,检测波长为280nm,柱温为35℃。结果:丁香酚在100.08~1 000.80μg/mL浓度范围内与其峰面积积分呈良好的线性关系,平均加样回收率为96.89%,RSD=1.01%。结论:建立的含量测定方法专属性强、准确度高、重复性好,可用于固本明目颗粒的质量控制。
Objective: To establish a HPLC method for the determination of eugenol in Guben Mingmu Granule Volatile Oil. Methods: The eugenol content was determined by high performance liquid chromatography (HPLC). The mobile phase consisted of Waters C18 (250 mm × 4.6 mm, 5 μm) and methanol-water (55:45, V / 1.0mL / min, the detection wavelength was 280nm, the column temperature was 35 ℃. Results: The eugenol showed a good linear relationship with the peak area in the range of 100.08-1 000.80 μg / mL. The average recovery was 96.89% and the RSD was 1.01%. Conclusion: The established method for the determination of content is highly specific, accurate and reproducible. It can be used for quality control of Guben Mingmu Granules.
