BURP是一类存在于植物中的特有蛋白,在生长发育和逆境响应等方面起重要作用。试验从青花菜中克隆到一个BURP基因,命名为Bo BURP1,在生物信息学分析的基础上,用RT-PCR研究了它在不同器官中的表达模式。研究结果表明,Bo BURP1的基因组DNA全长2 030 bp,具2个内含子;编码区全长为1 872bp,编码623个氨基酸;推导的蛋白质分子量为70.795 6 ku,等电点为8.65。系统发育分析结果表明,Bo BURP1与12种不同植物的同源蛋白可分为3组,Bo BURP1与十字花科植物同源序列的相似性最高,在进化树上聚为一组。RT-PCR结果表明,Bo BURP1在根、茎、叶、花蕾、花和角果中均有表达,叶、花蕾和花的表达量最高,而根和茎中的表达量最低。
BURP is a kind of unique protein existing in plants, which plays an important role in growth and development and adversity response. A BURP gene was cloned from broccoli and named Bo BURP1. Based on bioinformatics analysis, the expression pattern of BURP gene in different organs was studied by RT-PCR. The results showed that the full-length genomic DNA of Bo BURP1 was 2 030 bp with 2 introns. The full-length coding region of BURP1 was 1 872 bp encoding 623 amino acids. The deduced protein had a molecular weight of 70.795 6 ku and an isoelectric point of 8.65. Phylogenetic analysis showed that homologous proteins of Bo BURP1 and 12 different plants could be divided into three groups. Bo BURP1 had the highest similarity with homologous sequences of cruciferous plants, and clustered in a phylogenetic tree. RT-PCR results showed that Bo BURP1 was expressed in roots, stems, leaves, buds, flowers and pods, leaves, buds and flowers were the highest, while the lowest in roots and stems.
