为了研究牛磺鹅去氧胆酸(taurochenodeoxycholic acid,TCDCA)对佐剂性关节炎模型大鼠成纤维样滑膜细胞(fibroblast-like synoviocytes,FLS)中细胞间黏附分子-1(intercellular cell adhesion molecule-1,ICAM-1)表达的影响,采用qPCR与ELISA法检测了FLS中ICAM-1的表达量,结果表明,1×10~(-6)~1×10~(-4) mol/L TCDCA在基因与蛋白水平均能够显著或极显著抑制IL-1β刺激下FLS中ICAM-1的表达(P<0.05或P<0.01),且该抑制作用可被糖皮质激素受体(glucocorticoid receptor,GR)阻断剂米非司酮(RU486)拮抗,提示TCDCA对佐剂性关节炎的治疗作用与抑制GR活性进而抑制ICAM-1表达有关。
In order to study the effect of taurochenodeoxycholic acid (TCDCA) on the expression of intercellular cell adhesion molecule (VEGF) in fibroblast-like synoviocytes (FLS) 1, ICAM-1). The expression of ICAM-1 in FLS was detected by qPCR and ELISA. The results showed that the expression of ICAM-1 in FLS was significantly inhibited by 1 × 10 -6 ~ 1 × 10 -4 mol / L TCDCA at both gene and protein levels significantly or significantly inhibited ICAM-1 expression in FLS stimulated by IL-1β (P <0.05 or P <0.01), and the inhibitory effect could be inhibited by glucocorticoid receptor GR antagonist RU486, suggesting that the therapeutic effect of TCDCA on adjuvant arthritis is related to the inhibition of GR activity and the inhibition of ICAM-1 expression.
