目的:观察养心通脉片(YTT)对冠心病大鼠痰瘀痹阻证脂质代谢SREBP2通路基因m RNA的影响。方法:采用维生素D3和异丙肾上腺素复合高脂饲养建立大鼠心肌缺血痰瘀痹阻模型,随机分为3组。YTT组服YTT,绞股蓝总苷片(MET)组服MET,模型组不给药,另设正常大鼠为正常组。实验结束后检测血脂、血液流变学、用HE染色观察冠状动脉组织学变化,用逆转录聚合酶链反应(RT-PCR)法检测脂质代谢通路基因m RNA表达的变化。结果:YTT对冠心病大鼠有明显的降脂效果,能改善血液流变性,并能消退斑块,维持冠状动脉组织结构及功能,改善脂质代谢SREBP2通路基因m RNA的异常表达,使之趋于正常。结论:YTT具有良好的改善冠心病大鼠痰瘀病理的作用,它可能是通过调整脂质代谢SREBP2通路基因m RNA的表达来实现的。
Objective: To observe the effects of Yangxin Tongmai tablet (YTT) on m RNA of lipid metabolism SREBP2 pathway in phlegm and blood stasis syndrome of coronary heart disease rats. Methods: A rat model of myocardial ischemia and phlegm-stasis block was established by feeding high-fat vitamin D3 and isoproterenol combined with high-fat diet and randomly divided into 3 groups. YTT group served YTT, Gypenosides (MET) group served MET, the model group did not give the other set normal rats as the normal group. After the experiment, blood lipid and hemorheology were detected. Histological changes of coronary arteries were observed by HE staining. The mRNA expression of lipid metabolism pathway was detected by reverse transcriptase-polymerase chain reaction (RT-PCR). Results: YTT has obvious lipid-lowering effect on rats with coronary heart disease, can improve the blood rheology, and can eliminate plaque, maintain the structure and function of coronary artery tissue, and improve the expression of lipid metabolism SREBP2 pathway gene m RNA abnormal expression Tends to normal. CONCLUSION: YTT has a good effect on improving phlegm and blood stasis in rats with coronary heart disease, and it may be through regulating the expression of m RNA of lipid metabolism SREBP2 pathway gene.
