纯化的红花菜豆凝集素(PCL)经肼解、Bio-GelP-2柱分离,得到的寡糖部份用硼氢化钠还原,再经Bio-GelP-4柱层析,获得两条寡糖链。通过甲醇解、乙酰化反应,最后制成三甲基硅醚衍生物。气相色谱分析表明两个寡糖的组成为:Man8XylGlcNAc2(寡精I)和Man6XylGlcNAc2fuc(寡搪II)。用α-甘露糖苷酶、β-木糖苷酶、α-岩藻糖苷酶分别先后作用于两个寡糖,经酶解后的寡糖的大小在已标定的Bio-GelP-4柱上测定,从而得出两个寡糖的顺序分别为:(Manα),ManβXylβ(GlcNAc)2和(Manα)5 ManβXylβ(GlcNAc)2Fucα。
Purified Runner Bean Lectin (PCL) was hydrolyzed on a Bio-Gel P-2 column and the resulting oligosaccharide fraction was reduced with sodium borohydride and subjected to Bio-Gel P-4 column chromatography to obtain two oligosaccharides chain. Through methanolysis, acetylation reaction, and finally made of trimethylsilyl ether derivatives. Gas chromatographic analysis showed that the two oligosaccharides were composed of Man8XylGlcNAc2 (oligo I) and Man6XylGlcNAc2fuc (oligo II). The two oligosaccharides were treated with α-mannosidase, β-xylosidase and α-fucosidase respectively. The size of the oligosaccharides after enzymolysis was determined on a calibrated Bio-Gel P-4 column, The order of the two oligosaccharides was thus obtained as follows: (Manα), ManβXylβ (GlcNAc) 2 and (Manα) 5 ManβXylβ (GlcNAc) 2Fucα.
