为了解伪狂犬病病毒(PRV)和猪圆环病毒2型(PCV2)感染仔猪后不同时间点的病毒载量及排毒规律,将34头健康仔猪分为4组,第1~3组分别为人工接种PRV、PCV2和PRV+PCV2组,第4组为空白对照组。分别在接种后第3、7、14、21、35天采集试验仔猪的鼻拭子和肛门拭子,用Taq Man荧光定量PCR(q PCR)对其进行定量检测,并分析仔猪排毒的变化规律。结果,接种后第3天,3个试验组仔猪肛门拭子、鼻拭子中均分离到PRV或PCV2;其中PRV单独感染后的病毒载量在第14天达到峰值331.67 copies/L;PCV2单独感染后的病毒载量在第3天达到峰值10 742.01 copies/L;PRV+PCV2混合感染后鼻拭子和肛门拭子的PRV分别在第14天和第7天达峰值980.96 copies/L和2 230.056 copies/L;鼻拭子和肛门拭子PCV2在感染后第14天达到峰值69.413 copies/L和271.72 copies/L;混合感染仔猪于感染后第8~14天发生死亡。3个感染组之间无论是肛门拭子还是鼻拭子,PRV、PCV2病毒载量均差异显著(P<0.05)。上述结果表明,PRV单独感染、PCV2单独感染和二者混合感染均对仔猪造成严重危害,除引起仔猪发病死亡外,这2种病毒均能通过呼吸道和消化道向外排毒。
To understand the viral load and detoxification of piglets infected with PRV and PCV2 at different time points, 34 healthy piglets were divided into 4 groups. The first to third groups were artificial Inoculation of PRV, PCV2 and PRV + PCV2 group, the fourth group as a blank control group. Nasal swabs and anal swabs of piglets were collected on days 3, 7, 14, 21 and 35 after inoculation. Quantitative detection was performed by TaqMan quantitative PCR (qPCR) and the variation of detoxification . The results showed that PRV or PCV2 were isolated from anus swabs and nasal swabs on the third day after inoculation in piglets. The viral load after PRV infection alone reached a peak of 331.67 copies / L on day 14; PCV2 alone The viral load after infection peaked at 10 742.01 copies / L on day 3; PRV of nasal swab and anal swab reached a peak of 980.96 copies / L on day 14 and 7 after PRV + PCV2 mixed infection, respectively, and 2 230.056 copies / L; PCV2 of nasal swab and anal swab reached the peak of 69.413 copies / L and 271.72 copies / L on the 14th day after infection; and the piglets in mixed infection died on the 8th to 14th day after infection. The viral load of PRV and PCV2 were significantly different among the three infection groups (P <0.05), whether they were anal swabs or nasal swabs. The above results showed that PRV alone, PCV2 alone and mixed infection both caused serious damage to piglets. Except for the death of piglets, both viruses were able to detoxify through respiratory tract and digestive tract.
