目的 研究三七皂甙对严重体表烫伤大鼠心、肺组织Gia-mRNA表达的影响。方法 采用大鼠30%TBSAⅢ度烫伤模型,伤前20min给以不同剂量的三七皂甙,观察其对烫伤后3h心、肺组织Gi2α-,Gi3α-mRNA表达的影响;以半定量RT—PCR方法测定mRNA相对含量。结果 大鼠30%TBSAⅢ度烫伤后3h,Gi2α-mRNA含量在心肌增加178%(P<0.05),在肺组织增加263%(P<0.01);Gi3α-mRNA含量心肌增加236%(P<0.01),肺组织增加222%(P<0.05)。三七皂甙200mg/kg体重使烫伤Sh心肌Gi2α-mRNA含量减少46%(P<0.01);三七皂甙50、100、200mg/kg体重使烫伤3h肺组织Gi2α-mRNA含量分别减少36%、44%和61%(P<0.05或P<0.01),使烫伤3h心肌Gi3α-mRNA含量分别减少34%、30%和43%(P<0.05或P<0.01),使肺组织Ci3α-mRNA含量分别减少34%、55%和56%(P<0.05或P<0.01)。结论 三七皂甙对严重体表烫伤大鼠心肺组织Gj2α-,Gi3α-mRNA表达增加具有抑制作用。
Objective To study the effect of Panax notoginseng saponins on the expression of Gia-mRNA in heart and lung of severely burned rats. Methods Rats with 30% TBSA III degree scald model were given different doses of Panax notoginseng saponins at 20 min before injury to observe its effect on the expression of Gi2α- and Gi3α-mRNA in heart and lung tissues at 3 h after scald; semi-quantitative RT-PCR method was used. Determine the relative amount of mRNA. Results At 3 hours after 30% TBSA III degree scald in rats, Gi2α-mRNA increased by 178% (P<0.05) in the myocardium, increased by 263% in the lung tissue (P<0.01), and increased the myocardial content of Gi3α-mRNA by 236% (P<0.01). ) Lung tissue increased by 222% (P < 0.05). Panax Notoginseng 200mg/kg body weight reduced the myocardial Gi2α-mRNA content in the scalded Sh by 46% (P<0.01); Panax Notoginseng saponins 50, 100, and 200mg/kg body weight reduced the Gi2α-mRNA content in the lungs by 3% after scald for 3h, respectively. % and 61% (P<0.05 or P<0.01) decreased Gi3α-mRNA content in the myocardium at 3h after burn by 34%, 30% and 43%, respectively (P<0.05 or P<0.01), resulting in the expression of Ci3α mRNA in lung tissue. Decrease 34%, 55% and 56% (P<0.05 or P<0.01). Conclusion Panax Notoginseng has an inhibitory effect on the increase of Gj2α-, Gi3α-mRNA expression in heart and lung tissues of severely burned rats.
