鉴于目前常用的实验方法不够满意,作者试用尽可能与化妆品使用情况相似的两个方案来测定其过敏反应。一为开放性表皮诱导和激发,再用Freund氏佐剂(FCA)注入动物后腿以加强反应强度,二为通过类似皮内途径诱导以测定弱变应原的致敏潜力。选用的动物为Hartly albino豚鼠。每次实验用3组动物,即对照组,外用途经组和皮内组。每组用12或24只豚鼠,试验物有福马林、苯唑卡因,二硝基氯苯,对/邻/间甲基二氨基苯,对/邻/甲基氨基酚。试验用的化妆品处方为水包油型。试验用染料由Mrs.Bugaut实验室合成,A、B、C、D是用不同的脂族基置换的芳香胺。实验方法:1)外用途径诱导,所有动物第1天均用0.2mlFCA注入原脚垫。受试组于第1、3、5天以0.5ml试验物质涂
In view of the current unsatisfactory experimental methods used, the authors try to use as much as possible with the use of cosmetics two programs to determine the allergic reaction. One was induced by the open epidermis and challenged with Freund’s adjuvant (FCA) injected into the hind leg of the animal to enhance the reaction intensity. The other was induced by a similar intradermal route to determine the allergenic potential of the allergen. The animal used was Hartly albino guinea pig. Each experiment with three groups of animals, namely the control group, external use group and intradermal group. In each group, 12 or 24 guinea pigs were used. The test substance was fumarlin, benzocaine, dinitrochlorobenzene, para-o-m-methyldiaminobenzene and para-o-methylaminophenol. The cosmetic prescription for testing is oil-in-water. Test dyes were synthesized by Mrs.Bugaut Laboratories, and A, B, C and D are aromatic amines substituted with different aliphatic groups.
