目的探讨复方金铃四逆四物失笑散先期服药特色疗法治疗大鼠原发性痛经的镇痛机制。方法制备大鼠原发性痛经模型,采用先期服药法,用扭体法观察不同给药剂量对受试大鼠的镇痛效果,并考察受试药物对大鼠血浆中β-内啡肽(β-EP)含量及子宫内膜组织中前列腺素F2α(PGF2α)、前列腺素E2(PGE2)、6-酮-前列腺素F1α(6-keto-PGF1α)及血栓素B2(TXB2)含量的影响。结果复方金铃四逆四物失笑散各剂量对痛经大鼠均有明显镇痛效果,可显著减少30 min内大鼠的扭体次数,延长扭体潜伏期,镇痛作用有明显剂量相关性;可升高大鼠血浆中β-EP的含量,有效降低大鼠子宫组织中PGF2α、TXB2含量,明显升高PGE2和6-keto-PGF1α水平。结论复方金铃四逆四物失笑散在经前服药可达到治疗原发性痛经的效果,其作用机制可能与血浆中β-EP的含量,子宫内膜组织中PGF2α、PGE2、6-keto-PGF1α、TXB2的水平等一系列神经和内分泌的综合调节作用有关。
Objective To investigate the analgesic mechanism of Compound Jinling Sijin Si Liao San San for the treatment of primary dysmenorrhea. Methods The model of primary dysmenorrhea in rats was made. The analgesic effect of different doses on writhing rats was observed by writhing method. The effects of the drugs on rat plasma β-endorphin β-EP) and the levels of PGF2α, PGE2, 6-keto-PGF1α and TXB2 in endometrial tissue were determined. Results The compound Jinling Sijin Siqu Lixiao Powder had obvious analgesic effect on rats with dysmenorrhea, and significantly reduced the number of writhing in rats within 30 min, prolonging the incubation period of writhing, and the analgesic effect was obviously dose - dependent. Can increase the content of β-EP in rat plasma and effectively reduce the levels of PGF2α and TXB2 in rat uterine tissue and obviously increase the levels of PGE2 and 6-keto-PGF1α. Conclusions The compound Jinling Sijin Si Ni Si Xiao Liao San can be used in the treatment of primary dysmenorrhea. The mechanism may be related to the content of β-EP, the expression of PGF2α, PGE2, 6-keto-PGF1α in endometrium , TXB2 levels and a series of integrated regulation of nerve and endocrine role.
