目的观察和分析中链脂肪酸(辛酸/癸酸)对高脂膳食诱导的肥胖C57BL/6J小鼠小肠中TLR4传导通路相关分子表达的影响,探讨中链脂肪酸(MCFA)降体重的机制。方法 36只高脂饲料诱导的C57BL/6J肥胖小鼠按空腹体重随机分为3组,每组12只,分别给予含2%辛酸(C8:0)、癸酸(C10:0)和油酸(C18:1)的高脂饲料喂养16周,测定小鼠体重、肝脏重、肠系膜、附睾及肾周脂肪垫重,测定血清脂代谢指标,采用ELISA法测定小肠中肿瘤坏死因子α(TNF-α)、白介素-6(IL-6)、白介素-1β(IL-1β)水平,采用Real-time PCR法检测小肠组织中Toll样受体-4(TLR4)、髓样细胞分化因子88(My D88)和TNF-α的mRNA表达。结果辛酸和癸酸组肥胖小鼠体重、肝脏重、附睾周脂肪重、小肠组织中TNF-α水平以及TLR4 mRNA表达均显著低于油酸组(P<0.05)。辛酸组小鼠血清胆固醇(TC)和游离脂肪酸(FFA)、小肠组织中IL-6、IL-1β水平以及My D88、TNF-αmRNA表达显著低于油酸组(P<0.05),而高密度脂蛋白胆固醇(HDL-C)以及HDL-C/LDL-C显著高于油酸组(P<0.05)。癸酸组血清甘油三酯(TG)和FFA水平显著低于油酸组。结论 MCFA可能通过下调小肠中TLR4传导通路相关分子水平抑制肥胖个体炎症反应,降低肥胖个体体重。
Objective To observe and analyze the effect of medium chain fatty acids (caprylic / capric acid) on the expression of TLR4 pathway-related molecules in the small intestine of high fat diet-induced C57BL / 6J mice and to explore the mechanism of the weight loss of medium chain fatty acids (MCFA). Methods 36 high fat diet-induced C57BL / 6J obese mice were randomly divided into 3 groups according to their fasting body weight. Twelve mice in each group were given 2% octanoic acid (C8: 0), capric acid (C10: (C18: 1) high-fat diet for 16 weeks. The body weight, liver weight, mesentery, epididymis and perirenal fat pad weight of the mice were measured. Serum lipid metabolism indexes were measured. The levels of tumor necrosis factor- α, IL-6 and IL-1β were detected by real-time PCR. Toll-like receptor 4 (TLR4), myeloid differentiation factor 88 D88) and TNF-α mRNA expression. Results Body weight, liver weight, epididymal fat mass, TNF-α level and TLR4 mRNA expression in obese mice in octanoic acid and decanoic acid group were significantly lower than those in oleic acid group (P <0.05). Serum cholesterol (TC), free fatty acid (FFA) and IL-6, IL-1βlevels and My D88, TNF-αmRNA expressions in small intestine of octanoic acid group were significantly lower than those of oleic acid group (P <0.05) Lipoprotein cholesterol (HDL-C) and HDL-C / LDL-C were significantly higher than those in the oleic acid group (P <0.05). Serum triglyceride (TG) and FFA levels in capric acid group were significantly lower than those in oleic acid group. Conclusion MCFA may inhibit the inflammation of obese individuals and decrease the body weight of obese individuals by down-regulating TLR4-related molecular levels in the small intestine.
