目的探索β-连环蛋白在卵泡发育成熟过程中的表达及其作用。方法采用免疫组化法检测β-连环蛋白在3 w C57BL/6j小鼠卵巢中的表达及定位,进一步通过蛋白印迹技术检测β-连环蛋白在2 w、3 w、3 w经孕马血清预处理(3 w+PMSG 48 h)以及3 w经孕马血清及绒毛膜促性腺激素促排卵(3 w+PMSG 48 h+h CG 16 h)小鼠模型中的表达,观察其对卵泡发育的调控作用。结果免疫组化结果显示β-连环蛋白在3 w小鼠卵巢健康卵泡中广泛表达,而在闭锁卵泡中表达降低,其在始基卵泡、初级卵泡、小窦状卵泡中的表达与闭锁卵泡相比差异有统计学意义(P<0.05)。蛋白印迹检测结果显示其在3 w、3 w+PMSG 48 h、3 w+PMSG 48 h+h CG 16 h、2 w龄小鼠中的表达逐渐升高。结论β-连环蛋白表达随着卵泡发育成熟而降低,为进一步揭示卵巢衰老机制提供了新的研究方向。
Objective To explore the expression and role of β-catenin during follicular development and maturation. Methods The expression and localization of β-catenin in ovaries of 3-week-old C57BL / 6j mice were detected by immunohistochemistry. The levels of β-catenin in 2 w, 3 w and 3 w pregnant pregnant rabbits were detected by Western blotting (3 w + PMSG 48 h) and 3 w pregnant mare serum and chorionic gonadotropin ovulation (3 w + PMSG 48 h + h CG 16 h) mice model to observe the effect on follicular development Regulation. The result of Western blotting showed that the expression in the mouse at 3 w, 3 w + PMSG 48 h, 3 w + PMSG 48 h + h CG 16 h, 2 w was gradually increased. Conclusion The expression of β-catenin decreases with the development of ovarian follicles, which provides a new research direction for further revealing ovarian aging mechanism.
