目的:研究人参总皂苷对MAGE-12的B细胞表位抗体的增效作用。方法:将C-57BL/6小鼠随机分为对照组、MAP组、人参总皂苷组,MAP组在预测MAGE-12的B细胞表位序列为QSDEGSSNEEQE(82-93)的基础上,采用4分枝多重抗原肽结构设计合成抗原肽,免疫C-57BL/6小鼠,产生多克隆抗体,应用ELISA实验检测多克隆抗体是否为人的抗原肽的特异性抗体。人参总皂苷组在MAP抗原肽免疫小鼠的同时,人参总皂苷灌胃给药,0.2mL/20g,每日1次。结果:免疫C-57BL/6小鼠后可产生MAGE-12的抗原特异性的多克隆抗体,且人参总皂苷组抗体效价高于MAP组,达1∶256。结论:人参总皂苷对MAGE-12的B细胞表位抗体有增效作用。
Objective: To study the synergistic effect of ginsenosides on B cell epitopes of MAGE-12. METHODS: C-57BL/6 mice were randomly divided into control group, MAP group, and ginsenoside group. The MAP group was based on the prediction that the B cell epitope sequence of MAGE-12 was QSDEGSSNEEQE (82-93). Branching multiple antigens peptide structure Design and synthesis of antigenic peptides. C-57BL/6 mice were immunized to produce polyclonal antibodies. ELISA was used to test whether the polyclonal antibodies were specific antibodies to human antigenic peptides. Ginseng total saponin group was administrated with ginseng total saponin by gavage at the same time as the MAP antigen peptide was immunized to mice, 0.2 mL/20 g once a day. RESULTS: After immunization with C-57BL/6 mice, antigen-specific polyclonal antibodies against MAGE-12 were produced, and the ginsenosides antibody titer was higher than that of the MAP group, reaching 1:256. CONCLUSION: Ginsenosides have a synergistic effect on B cell epitopes of MAGE-12.
