目的建立测定人血浆中依托考昔浓度的高效液相色谱法。方法以150 mm×4.6 mm,3.5μm(Waters xbridge)为色谱柱,流动相为水∶乙腈(58∶42),流速为1 ml/min,检测波长283 nm,以乙腈沉淀提取。结果依托考昔高、中、低(0.9375μg/ml,60μg/ml,100μg/ml)3种浓度的平均回收率分别为84.59%,96.89%,82.85%,日内、日间差RSD均低于6%;分析方法的最低检测浓度为0.5μg/ml,线性范围为0.5μg/ml~120μg/ml。结论该方法灵敏、准确、快速、简单,可用于临床血药浓度监测和药动学研究。
Objective To establish a HPLC method for the determination of etoposide in human plasma. Methods The mobile phase consisted of water: acetonitrile (58:42) at a flow rate of 1 ml / min and a detection wavelength of 283 nm on a 150 × 4.6 mm, 3.5 μm (Waters xbridge) column. Results The average recoveries of three concentrations of propoxly, middle and low (0.9375μg / ml, 60μg / ml, 100μg / ml) were 84.59%, 96.89% and 82.85% respectively. 6%; the lowest detection concentration of the analytical method was 0.5μg / ml, the linear range of 0.5μg / ml ~ 120μg / ml. Conclusion The method is sensitive, accurate, rapid and simple and can be used for clinical blood drug concentration monitoring and pharmacokinetic studies.
