目的运用基因芯片技术,观察二乙基亚硝胺(DEN)诱导大鼠肝癌(含肝)组织与正常对照大鼠肝组织比较表达下调的基因。方法DEN诱发大鼠肝癌,应用AffymetrixRat230AGeneChip进行检测。结果在芯片的15710个基因中,剔除正常肝组织表达值读数在50以下的基因,正常肝脏基因表达大于肝癌基因的有248个,其中120个为EST片段,已知基因为128个。结论DEN诱发大鼠肝癌的后基因组变化是十分复杂的,如何逐渐找到那些起着关键作用的基因,进一步阐释其在肝癌的发生、发展和转归中的作用,是今后工作的重点。
OBJECTIVE: To study the down-regulation genes of rat liver cancer (including liver) and normal control rats induced by diethylnitrosamine (DEN) using gene chip technology. Methods DEN-induced rat hepatocellular carcinoma was detected by Affymetrix Rat230 AGeneChip. Results Among 15710 genes in the chip, 248 genes whose expression in normal liver were less than 50 were excluded from expression of normal liver tissue, of which 120 were EST fragments and 128 were known genes. Conclusions The postnatal change of DEN-induced hepatocarcinoma in rats is very complicated. How to gradually find those genes that play a key role in further explaining their roles in the occurrence, development and prognosis of hepatocellular carcinoma is the focus of future work.
