采用RNA斑点杂交分析方法,对40例食管癌组织和相对应的正常粘膜组织中谷胱甘肽硫转移酶(GlutathioneStransferase,GSTs)同功酶基因GSTπ、GSTα和GSTμ基因的表达情况进行研究,结果食管癌组织中GSTπ基因表达阳性率为77.5%,在相应的正常粘膜组织中表达的阳性率为90.0%。在食管癌组织中GSTα和GSTμ基因表达水平较低,表达阳性率分别为35.0%和20.0%,而正常粘膜组织中GSTα和GSTμ基因表达的阳性率分别为20%和15%,GSTs在癌组织中表达阳性率与正常粘膜组织相比,P>0.05。GSTπ基因表达阳性率明显高于GSTα和GSTμ基因,P<0.05。结果提示:GSTs同功酶基因在食管癌发生发展中基因转录水平增高,GSTπ是食管癌中GSTs同功酶存在的主要形式。
The glutathione S-transferase (GSTs) isoenzyme gene GST , GSTα, and GSTμ genes in 40 esophageal cancer tissues and corresponding normal mucosal tissues were analyzed by RNA dot blot hybridization. The expression status of GST-π was found to be 77.5% in esophageal cancer tissues and 90.0% in corresponding normal mucosa tissues. The expression of GSTα and GSTμgenes was low in esophageal cancer tissues, and the positive rates were 35.0% and 20.0%, respectively. The positive rates of GSTα and GSTμgene expression in normal mucosal tissues The positive rates of GSTs in cancer tissues were 20% and 15%, respectively, compared with normal mucosal tissues, P>0.05. The positive rate of GSTπ gene expression was significantly higher than that of GSTα and GSTμ, P<0.05. The results suggest that the gene transcription level of GSTs isoenzyme gene in the development of esophageal cancer is increased, GST-π is the main form of GSTs isoenzyme in esophageal cancer.
