以草莓(Fragaria×ananassa)品种‘花姬’为试材,通过PCR和RACE的方法克隆出LFY同源基因的cDNA全长序列,命名为FaLFY(GenBank收录号JN788260),通过定量PCR的方法检测了该基因在草莓植株各组织和花器官中的表达。结果表明FaLFY编码区长度为1236bp,编码412个氨基酸,与其他物种的氨基酸序列都有一定的同源性,其中与蔷薇科玫瑰的LEAFY-1同源性最高,达到88%。实时定量RT-PCR结果表明,在不同的组织、不同的花器官中FaLFY的表达量存在差异,其中在花芽中的表达量最高,在营养生长的组织中基本不表达,在成熟花器官中同样不表达,说明该基因在草莓的成花进程中发挥重要作用。
The full-length cDNA sequence of LFY homologous gene was cloned by PCR and RACE, and was named FaLFY (GenBank Accession No. JN788260) by Fragaria × ananassa cultivar ’Daji’. The quantitative PCR was used to detect The expression of this gene in various tissues and flower organs of strawberry plants was studied. The results showed that the FaLFY coding region was 1236 bp in length and encoded a polypeptide of 412 amino acids, which shared some homology with the other species. The highest homology was 88% with LEAFY-1 of Rosaceae rose. The results of real-time quantitative RT-PCR showed that the FaLFY expression levels were different in different tissues and floral organs. Among them, the expression level of FaLFY was the highest in flower buds, but not in vegetative tissues. In mature flower organs, No expression, indicating that the gene plays an important role in strawberry flower process.
