目的研究血小板源性生长因子BB(PDGF-BB)对人卵巢癌微血管内皮细胞(OdMEC)增殖、增殖细胞核抗原(PC- NA)和血管内皮生长因子(VEGF)表达的影响。方法体外分离培养OdMEC,施加不同浓度的PDGF-BB(0．5．10．20．50ng／ml),采用四甲基偶氮唑(MTT)法测定细胞生长曲线；免疫细胞化学及RT-PCR方法检测PCNA和VEGF表达的变化。结果PDGF-BB能促进OdMEC的增殖,呈剂量依赖性,以20ng／ml最为明显(P＜0．05)。在20ng／mI PDGF-BB刺激下,PCNA和VEGF的表达均高于时照组,差异有统计学意义(P＜0．05)。结论PDGF-BB促进OdMEC的增殖、PCNA和VEGF的表达。
Objective To investigate the effects of platelet derived growth factor BB (PDGF-BB) on the proliferation, the expression of proliferating cell nuclear antigen (PCNA) and vascular endothelial growth factor (VEGF) in human ovarian cancer microvascular endothelial cells (OdMECs) Methods OdMEC was isolated and cultured in vitro. Different concentrations of PDGF-BB (0.5.10.20.50ng / ml) were added and the cell growth curve was determined by MTT assay. Immunocytochemistry and RT-PCR Methods The changes of PCNA and VEGF expression were detected. Results PDGF-BB could promote the proliferation of OdMEC in a dose-dependent manner, most obviously at 20 ng / ml (P <0.05). The expression of PCNA and VEGF in PDGF-BB stimulated by 20ng / mI were significantly higher than those in control group (P <0.05). Conclusion PDGF-BB can promote the proliferation of OdMEC, the expression of PCNA and VEGF.
