目的调查山东省济宁地区人群16个Y-STR基因座及其单倍型的遗传多态性,探讨其法学应用价值,为法医学应用提供基础数据。方法应用AmpFlSTR Yfiler试剂盒,对济宁地区197名汉族无关男性个体进行16个Y-STR基因座的复合扩增,用ABI3130型基因分析仪对扩增产物进行检测,统计其群体遗传学参数。结果前15个基因座分别检出6、5、6、6、8、6、6、3、7、8、6、4、3、4、6个等位基因。DYS385 a检出10个等位基因,DYS385 b有165例表达,检出14个等位基因,DYS385 a/b基因座检出47种单倍型。基因多样性(GD)值最低为0.3867(DYS438),最高为0.9717(DYS385 a/b)。在165例全表达对象中,观察到16个Y-STR基因座共同构成的单倍型160种,其中159种单倍型只出现1次,5种出现2次,1种出现3次,累计GD值为0.9994。结论 16个Y-STR基因座具有较强的个体识别能力,可应用于法庭科学中的个体识别与亲权鉴定。
Objective To investigate the genetic polymorphisms of 16 Y-STR loci and their haplotypes in the population of Jining, Shandong Province, and to explore the value of their application in law and to provide basic data for forensic applications. Methods AmpFlSTR Yfiler kit was used to amplify 16 Y-STR loci in 197 Han unrelated male individuals in Jining. The amplified products were detected by ABI3130 gene analyzer and the population genetic parameters were calculated. Results The first 15 loci were detected 6,5,6,6,8,6,6,3,7,8,6,4,3,4,6 alleles. DYS385 a detected 10 alleles, DYS385 b 165 expressed, detected 14 alleles, DYS385 a / b loci detected 47 haplotypes. The lowest GD was 0.3867 (DYS438) and the highest was 0.9717 (DYS385 a / b). Of the 165 fully expressed objects, 160 haplotypes were found in 16 Y-STR loci, of which 159 haplotypes appeared only once, five appeared twice, one appeared three times, and accumulated The GD value is 0.9994. Conclusion The 16 Y-STR loci have strong individual identification ability and can be applied to individual identification and paternity testing in forensic science.
