目的:探讨中药醒脑液(XNY)对PAP双转基因痴呆小鼠大脑组织中中性内肽酶(NEP)基因和蛋白表达的影响。方法:分别采用实时荧光定量PCR技术和Western blot技术观测正常对照组、模型组、西药对照组、XNY高剂量组、XNY中剂量组和XNY低剂量组小鼠大脑组织中NEPmRNA和NEP蛋白的表达。结果:与模型组相比,XNY高剂量组脑内NEP mRNA含量表达上调(P<0.01);XNY高剂量组和中剂量组NEP蛋白含量表达增加(P<0.01)。结论:醒脑液对双转基因痴呆小鼠大脑组织中NEP mRNA和NEP蛋白表达有较好的调控作用,说明醒脑液对防治老年性痴呆能起到良好效果。
Objective: To investigate the effect of xingyin liquid (XNY) on the expression of neutral endopeptidase (NEP) gene and protein in the brain of PAP double transgenic dementia mice. Methods: The expression of NEP mRNA and NEP in brain tissues of mice in normal control group, model group, western medicine control group, XNY high dose group, XNY middle dose group and XNY low dose group were observed by real-time fluorescence quantitative PCR and Western blot respectively . Results: Compared with the model group, the expression of NEP mRNA in high dose XNY group was increased (P <0.01). The expression of NEP protein in high dose XNY group and medium dose group was increased (P <0.01). CONCLUSION: Xingnao liquid has good regulation on the expression of NEP mRNA and NEP protein in the brain of double transgenic dementia mice, indicating that Xingnao liquid can play a good role in the prevention and treatment of senile dementia.
