目的:研究白藜芦醇对胃癌细胞株增殖和凋亡的影响。方法:不同浓度白藜芦醇(0~150μmol/L)作用人胃腺癌细胞株BGC823(低分化)和SGC7901(中分化),CCK-8试剂盒检测细胞增殖,流式细胞仪检测细胞凋亡,免疫印迹检测凋亡相关蛋白survivin的表达。结果:白藜芦醇对胃癌细胞株增殖能力呈时间和浓度依赖性抑制,150μmol/L白藜芦醇作用人胃癌细胞株BGC823和SGC7901 72 h后,早期和晚期凋亡细胞比率均明显增加,而survivin蛋白表达均明显下降(P<0.05)。结论:白藜芦醇以浓度和时间依赖性模式抑制胃癌细胞的增殖能力和活性,诱导胃癌细胞株BGC823和SGC7901发生凋亡,其机制可能与下调survivin蛋白的表达有关。
Objective: To study the effects of resveratrol on proliferation and apoptosis of gastric cancer cell lines. Methods: Human gastric adenocarcinoma cell line BGC823 (low differentiation) and SGC7901 (moderately differentiated) were treated with different concentrations of resveratrol (0-150 μmol / L). Cell proliferation was detected by CCK-8 kit and apoptosis was detected by flow cytometry Western blot was used to detect the expression of survivin. Results: Resveratrol inhibited the proliferation of gastric cancer cells in a time and concentration-dependent manner. After treated with 150 μmol / L resveratrol for 72 h, the rates of apoptotic cells in both early and late stages were significantly increased, While the expression of survivin protein decreased significantly (P <0.05). CONCLUSION: Resveratrol inhibits the proliferation and activity of gastric cancer cells in a concentration-dependent and time-dependent manner and induces the apoptosis of gastric cancer cell lines BGC823 and SGC7901. The mechanism may be related to the downregulation of survivin protein.
