目的寻找过路黄(Lysimachia christinae Hance)抗肿瘤活性成分。方法采用AB-8大孔吸附树脂色谱、硅胶层析柱色谱、Sephadex LH-20凝胶柱色谱、ODS柱色谱和制备HPLC等技术方法进行化学成分分离,根据理化性质和波谱数据鉴定化合物结构。MTT法检测化合物体外抗肿瘤细胞生长活性。结果从过路黄干燥全草70%乙醇提取物中分离得到11个化合物,分别鉴定为:3,3’-二甲氧基-6,6’-双[(Z)-十五烷-10-烯-1-基)]-(1,1’-双环己烷)-3,3’,6,6’-四烯-2,2’,5,5’-四酮(1),大黄素甲醚(2),茜草色素(3),9,16-二羰基-10,12,14-三烯十八碳酸(4),西克拉敏A糖苷A-3-O-{β-D-吡喃木糖基-(1→2)-β-D-吡喃葡萄糖基-(1→2)-[β-D-吡喃葡萄糖基-(1→4)]-α-L-吡喃阿拉伯糖苷}(5),原报春花素A-3-O{β-D-吡喃木糖基-(1→2)-β-D-吡喃葡萄糖基-(1→4)-[β-D-吡喃葡萄糖基-(1→2)]-α-L-吡喃阿拉伯糖苷}(6),槲皮素-3-O-吡喃葡萄糖苷(7),异鼠李素-3-O-β-D-吡喃半乳糖苷(8),3’,4’,5’,5,7-五羟基黄酮(9),芦丁(10),槲皮素(11)。化合物5对人宫颈癌细胞株He La、人骨肉瘤细胞株U20S、人肺腺癌细胞株PC-9、人结肠癌细胞株CT-26的生长均有一定的抑制作用,其IC50值分别为2.29、1.22、1.43、1.29μmol/L。结论化合物1为新化合物,化合物2~4为首次从该属植物中分离得到。化合物5在体外具有显著抗肿瘤细胞生长活性。
Objective To search for anti-tumor active ingredients of Lysimachia christinae Hance. Methods The chemical constituents were isolated by AB-8 macroporous adsorption resin, silica gel column chromatography, Sephadex LH-20 gel column chromatography, ODS column chromatography and preparative HPLC. The structures of the compounds were identified by physico-chemical properties and spectral data. The anti-tumor cell growth activity of the compounds was tested by MTT assay. Results Eleven compounds were isolated from the 70% ethanol extract of the whole plant, and were identified as 3,3’-dimethoxy-6,6’-bis [(Z) -pentadecane- (1,1’-dicyclohexyl) -3,3 ’, 6,6’-tetraene-2,2’, 5,5’-tetraone (1), emodin Methyl ether (2), madder pigment (3), 9,16-dicarbonyl-10,12,14-triene octadecaic acid (4), ciclantiminoside A-3-O- { (1 → 2) -β-D-glucopyranosyl- (1 → 2) - [β-D-glucopyranosyl- (1 → 4)] - α-L-pyran (1 → 2) -β-D-glucopyranosyl- (1 → 4) - [β-D-glucopyranosyl- (1 → 2)] - α-L-arabinopyranoside} (6), quercetin-3-O-glucopyranoside (7), isorhamnetin-3 -O-β-D-galactopyranoside (8), 3 ’, 4’, 5 ’, 5,7-pentahydroxyflavone (9), rutin (10) and quercetin (11). Compound 5 has certain inhibitory effects on the growth of human cervical cancer cell line He La, human osteosarcoma cell line U20S, human lung adenocarcinoma cell line PC-9 and human colon cancer cell line CT-26, with IC50 values of 2.29 , 1.22, 1.43, 1.29 μmol / L. Conclusion Compound 1 is a new compound, and compounds 2 ~ 4 are isolated from this genus for the first time. Compound 5 has significant anti-tumor cell growth activity in vitro.
