目的研究nodosin对大鼠胶质瘤C6细胞的抗肿瘤作用。方法采用倒置显微镜对C6细胞进行形态学观察;采用四甲基偶氮唑蓝(MTT)法测定细胞生存率,乳酸脱氢酶(LDH)法检测细胞毒性,BrdU检测法检测细胞增殖情况;采用Hoechst 33342染色法观察nodosin对C6细胞凋亡的影响。结果 Nodosin在作用12 h和24 h可抑制大鼠胶质瘤C6细胞的细胞生存率,呈时间与剂量依赖性;在0~20μmol.L-1浓度范围内对C6细胞无细胞毒性作用;20μmol.L-1nodosin作用24 h,C6细胞增殖率降低至74.6%;同时Nodosin能够促进C6细胞的凋亡。结论 Nodosin具有抗肿瘤作用,其作用与抑制细胞的增殖及促进细胞的凋亡有关。
Objective To study the antitumor effect of nodosin on rat C6 glioma cells. Methods The morphology of C6 cells was observed by inverted microscope. The cell viability was determined by MTT assay, the cytotoxicity by lactate dehydrogenase (LDH) assay and the cell proliferation by BrdU assay. Hoechst 33342 staining was used to observe the effect of nodosin on C6 cell apoptosis. Results Nodosin inhibited the cell viability of rat C6 glioma cells in a dose-and time-dependent manner at 12 h and 24 h. No cytotoxic effects were found on C6 cells at a concentration of 0 ~ 20 μmol·L-1. The proliferation rate of C6 cells decreased to 74.6% after L-1nodosin treatment for 24 h, while Nodosin could promote the apoptosis of C6 cells. Conclusion Nodosin has anti-tumor effect, and its role in inhibiting cell proliferation and promoting cell apoptosis.
