目的检测循环miR-128在结直肠癌患者血清中的表达,观察其对结直肠癌细胞迁移侵袭能力的影响。方法选择2012年6月至12月在山东大学齐鲁医院行结直肠癌根治术的结直肠癌患者57例(结直肠癌组,又分为转移组和未转移组),以健康查体的志愿者20例(健康对照组)为参照。实时荧光定量PCR法检测两组血清中miR-128的表达,分析其与结直肠癌转移的关系;采用Lipofectamine 2000将miR-128模拟物转染入HT-29细胞,观察miR-128过表达对HT-29细胞迁移侵袭能力的影响。结果结直肠癌组血清miR-128水平明显低于健康对照组(P<0.001),其中转移组水平明显低于未转移组(P=0.014)。ROC曲线分析显示,血清miR-128对诊断结直肠癌和转移鉴别诊断的效能分别为0.85、0.71(P均<0.05)。miR-128过表达使HT-29细胞迁移愈合率以及迁移、侵袭细胞数明显减少。结论循环miR-128在结直肠癌患者血清中表达明显下调,并且与转移密切相关,为结直肠癌转移诊断和治疗提供了新思路。
Objective To detect the expression of circulating miR-128 in the serum of patients with colorectal cancer and to observe its effect on the migration and invasion of colorectal cancer cells. Methods 57 patients with colorectal cancer (colorectal cancer group, divided into metastasis group and non-metastasis group) who underwent radical resection of colorectal cancer in Qilu Hospital of Shandong University from June to December 2012 were enrolled in this study. 20 cases (healthy control group) as a reference. The expression of miR-128 in sera of two groups was detected by real-time fluorescence quantitative PCR, and its relationship with metastasis of colorectal cancer was analyzed. The miR-128 mimics were transfected into HT-29 cells by Lipofectamine 2000 to observe the expression of miR-128 overexpression HT-29 cell migration and invasion. Results The level of miR-128 in colorectal cancer group was significantly lower than that in healthy control group (P <0.001). The level of miR-128 in metastasis group was significantly lower than that in non-metastasis group (P = 0.014). ROC curve analysis showed that the efficacy of serum miR-128 in the differential diagnosis of colorectal cancer and metastasis was 0.85 and 0.71, respectively (all P <0.05). MiR-128 overexpression resulted in a significant decrease in the number of migrating cells and migration of HT-29 cells. Conclusion The expression of circulating miR-128 is significantly down-regulated in the serum of patients with colorectal cancer and is closely related to metastasis. It provides a new idea for the diagnosis and treatment of metastatic colorectal cancer.
