[目的]探讨人骨髓核分化抗原(MNDA)对人骨肉瘤Saos-2细胞增殖及侵袭的影响。[方法]逆转录聚合酶链反应(RT-PCR)法定量测定Saos-2细胞中MNDA的mRNA表达差异。将MNDA基因转染Saos-2细胞,MTT法测定检测细胞增殖程度的改变,Transwell小室法测定肿瘤细胞侵袭能力的改变。没有转染质粒的和转染空质粒的Saos-2做为对照组。[结果]MNDA的mRNA在骨肉瘤Saos-2细胞中表达明显降低。转染MNDA的人骨肉瘤Saos-2细胞的增殖率低于对照组,并且抑制能力随时间延长抑制而增加,侵袭穿透能力减弱。[结论]人骨肉瘤转染MNDA可抑制肿瘤细胞的增殖,降低肿瘤细胞的侵袭穿透能力。
[Objective] To investigate the effect of human bone marrow nuclear antigen (MNDA) on the proliferation and invasion of human osteosarcoma Saos-2 cells. [Method] The mRNA expression of MNDA in Saos-2 cells was quantitatively determined by reverse transcription-polymerase chain reaction (RT-PCR). The MNDA gene was transfected into Saos-2 cells. The cell proliferation was detected by MTT assay. The invasion ability of tumor cells was determined by Transwell chamber assay. Saos-2 without plasmid transfection and transfected with empty plasmid served as control group. [Results] The mRNA expression of MNDA was significantly decreased in osteosarcoma Saos-2 cells. The proliferation rate of MNOS-transfected human osteosarcoma Saos-2 cells was lower than that of the control group, and the inhibitory ability of MNOS transfected with MNDA increased with the prolongation of inhibition and the invasion and penetration ability weakened. [Conclusion] MNDA transfected with human osteosarcoma can inhibit the proliferation of tumor cells and reduce the invasion and penetration of tumor cells.
