通过正相硅胶、Sephadex LH-20和反相HPLC色谱分离方法相结合,从华南木姜子树枝乙醇提取物的乙酸乙酯部位中分离得到1个新苯乙烯二聚体衍生物(1),经IR,UV,MS和NMR(1D和2D NMR)波谱学方法鉴定为(E)-2,4-双(对-羟基苯基)-2-丁烯醇[(E)-2,4-bis(p-hydroxyphenyl)-2-butenol],命名为木姜子醇(listeanol)。在1×10-5mol·L-1浓度下,它在肿瘤细胞毒(MTT法,HCT-8,Bel-7402,BGC-823,A549,A2780细胞株)、抗氧化(Fe2+_Cys诱导大鼠肝微粒体脂质过氧化模型)、神经保护(去血清和谷氨酸损伤模型)和抗糖尿病(PTPIB酶抑制模型)体外模型上均未显示出显著药理活性。
A novel styrene dimer derivative (1) was isolated from the ethyl acetate fraction of the ethanolic extract of A. annua through normal phase silica gel, Sephadex LH-20 and reversed-phase HPLC chromatographic separation. (E) -2,4-bis (p-hydroxyphenyl) -2-butenol was identified by IR, UV, MS and NMR spectroscopic methods (1D and 2D NMR) bis (p-hydroxyphenyl) -2-butenol], named listeanol. The anti-oxidant (Fe2 + _Cys-induced rat) cells were cultured in the presence of 1 × 10-5 mol·L-1 of tumor cell cytotoxicity (MTT method, HCT-8, Bel 7402, BGC-823, A549 and A2780 cell lines) Liver microsomal lipid peroxidation model), neuroprotection (de-serum and glutamate damage models) and anti-diabetic (PTPIB enzyme inhibition model) in vitro models showed no significant pharmacological activity.
