本文报道自肝细胞癌病人的手术切除标本建立的一个人体肝癌细胞系,YY-8103。经形态,超微结构观察,染色体分析,乳酸脱氢酶(LDH)同工酶谱测定,刀豆球蛋白A(Con.A)凝集试验和甲胎蛋白(AFP)检测,确定该细胞系为人体肝癌细胞,至今已传了130多代。该细胞容易接种于裸鼠,而且其瘤结节能在体外再培养。体外培养时该细胞不具有合成乙型肝炎表面抗原(HBsAg)的功能,但对麻疹病毒及单纯疱疹病毒敏感。因而认为在肝癌基础研究和病毒试验中可得到应用。
This article reports a human hepatocellular carcinoma cell line, YY-8103, established from surgical specimens of patients with hepatocellular carcinoma. The morphology, ultrastructure observation, chromosome analysis, determination of lactate dehydrogenase (LDH) isozyme zymography, concanavalin A (Con.A) agglutination test, and alpha fetoprotein (AFP) assay were used to determine the cell line as Human liver cancer cells have been transmitted for more than 130 generations so far. The cells were easily inoculated in nude mice, and their tumor nodes were energy-saving and re-incubated in vitro. When cultured in vitro, the cells do not have the function of synthesis of hepatitis B surface antigen (HBsAg) but are sensitive to measles virus and herpes simplex virus. Therefore, it is considered that it can be applied in basic research and virus tests of liver cancer.
