研究芍药苷、薄荷脑对葛根素在细胞模型转运过程中对Calu-3细胞膜生理功能的影响。以Calu-3细胞作为模拟鼻黏膜组织的体外细胞模型,采用荧光漂白恢复技术及超微量酶活性检测方法,考察细胞膜流动性、Na~+-K~+-ATP酶和Ca~(2+)-ATP酶活性,探索配伍药物促进葛根素转运的作用机制。结果表明,配伍低、中、高浓度薄荷脑或同时配伍低、中、高浓度芍药苷和薄荷脑时,与单独使用葛根素相比,Calu-3细胞膜荧光漂白恢复率显著升高,Na~+-K~+-ATP酶活性未发生明显改变,Ca~(2+)-ATP酶活性显著增强,由此证实薄荷脑起到主要渗透促进作用,并且其作用机制可能与增加流动性、激活Ca~(2+)-ATP酶活性密切相关。
To study the effect of paeoniflorin and menthol on the physiological function of Calu-3 cell membrane during the transport of puerarin. Using Calu-3 cells as a model of nasal mucosa in vitro, the cell membrane fluidity, Na ~ + -K ~ + -ATPase and Ca ~ (2+) were investigated by fluorescence bleaching recovery assay and ultra- -ATP enzyme activity to explore the role of compatibility drugs to promote puerarin transport mechanism. The results showed that when compared with low, medium and high concentrations of menthol or with low, medium and high concentrations of paeoniflorin and menthol, the fluorescence recovery of Calu-3 cells was significantly increased compared with puerarin alone. Na ~ + -K ~ + -ATP activity did not change significantly, Ca ~ (2 +) - ATPase activity was significantly enhanced, which confirmed that menthol play a major role in promoting penetration, and its mechanism may increase mobility, activation Ca ~ (2 +) - ATPase activity is closely related.
