目的建立氯霉素滴眼液中硫柳汞和苯基汞防腐剂的含量测定方法。方法采用高效液相色谱法,色谱柱为Agilent苯基柱[Zorbax Eclipse XDB-phenyl(4.6 mm×250 mm,5μm)],0.05 mol·L-1磷酸二氢钾溶液(用三乙胺调节pH值至5.5)-乙腈(82∶18)为流动相,流速为1.2 mL·min-1,检测波长为220 nm。结果硫柳汞和醋酸苯汞在2~100μg·mL-1之间呈良好的线性关系(r>0.999 0),硫柳汞的平均回收率≥96.9%,醋酸苯汞的平均回收率≥99.1%。结论本法简便、准确、专属性强,可用于药品中有机汞防腐剂的控制。
Objective To establish a method for the determination of thimerosal and phenylmercury preservatives in chloramphenicol eye drops. Methods High performance liquid chromatography (HPLC) was used. The column was Agilent Phenyl column [Zorbax Eclipse XDB-phenyl (4.6 mm × 250 mm, 5 μm)], 0.05 mol·L -1 potassium dihydrogen phosphate solution Value to 5.5) -acetonitrile (82:18) as mobile phase with a flow rate of 1.2 mL · min-1 and a detection wavelength of 220 nm. Results The results showed that thimerosal and phenylmercuric acetate showed a good linearity (r> 0.999 0) between 2 and 100 μg · mL-1. The average recovery of thimerosal was 96.9% and the average recovery of phenylmercuric acetate was 99.1%. Conclusion This method is simple, accurate and specific and can be used for the control of organic mercury preservatives in medicines.
