目的探讨双歧杆菌对过敏性哮喘儿童外周血单核细胞(PBMC)来源的树突状细胞(DC)分泌IL-1β、IL-6、IL-10、IL-12、IL-23和IFN-γ的影响。方法从15例过敏性哮喘儿童和15例非哮喘儿童的外周血单个核细胞诱导生成未成熟DC,加入双歧杆菌后继续培养DC 2 d,用ELISA方法检测培养上清中IL-1β、IL-6、IL-10、IL-12、IL-23和IFN-γ的水平。结果双歧杆菌能明显刺激哮喘儿童DC分泌IL-12、IFN-γ、IL-1β及IL-6和非哮喘儿童DC分泌IL-12、IL-10、IL-1β及IL-23水平增高。结论双歧杆菌能够刺激过敏性哮喘儿童DC分泌IL-12和IFN-γ,可能改变Th2优势分化,纠正Th1/Th2失衡。同时双歧杆菌还能刺激哮喘儿童DC分泌IL-1β及IL-6增高,达到促进Th17细胞分化的作用。
Objective To investigate the effects of Bifidobacterium on the secretion of IL-1β, IL-6, IL-10, IL-12, IL-23 and IFN-γ by dendritic cells derived from peripheral blood mononuclear cells (PBMCs) γ effect. Methods Immature DCs were induced from peripheral blood mononuclear cells from 15 allergic asthmatic children and 15 non-asthmatic children. After adding Bifidobacterium, DCs were continuously cultured for 2 days. The levels of IL-1β, IL -6, IL-10, IL-12, IL-23 and IFN-γ. Results Bifidobacterium significantly increased the levels of IL-12, IL-10, IL-1β and IL-23 secreted by DCs secreted by DC from DCs, IL-12, IL-1β, IL-6 and non-asthmatic children. Conclusions Bifidobacteria can stimulate the secretion of IL-12 and IFN-γ by DC in children with allergic asthma, which may change the dominant differentiation of Th2 and correct the imbalance of Th1 / Th2. Bifidobacteria can also stimulate the secretion of IL-1β and IL-6 in DCs of asthmatic children, and promote the differentiation of Th17 cells.
