目的建立高效液相色谱测定多索茶碱血药浓度的方法。方法用乙醚提取血样,以卡马西平为内标进行检测。色谱柱采用Waters C18(3.9 mm×150 mm,5μm),流动相为甲醇-水(35∶65),流速为0.6 mL·min-1,检测波长为273 nm,柱温为30℃。结果多索茶碱血清浓度在0.7~28μg·mL-1内线性关系良好,线性方程为Y=0.354 9X+0.003 9(r=0.999 7),方法回收率为100.4%,日内、日间RSD分别为1.10%~3.87%和2.11%~4.86%(n=5)。结论该方法快速、简便、准确、安全,可用于临床多索茶碱血药浓度的测定。
Objective To establish a HPLC method for the determination of doxofylline plasma concentration. Methods Blood samples were extracted with ether and detected with carbamazepine as internal standard. The mobile phase consisted of methanol-water (35:65), the flow rate was 0.6 mL · min-1, the detection wavelength was 273 nm and the column temperature was 30 ℃. The chromatographic column was Waters C18 (3.9 mm × 150 mm, 5 μm). Results The doxofylline had good linearity in the range of 0.7 ~ 28μg · mL-1. The linear equation was Y = 0.354 9X + 0.0039 (r = 0.999 7). The recovery rate was 100.4% 1.10% ~ 3.87% and 2.11% ~ 4.86% (n = 5). Conclusion The method is rapid, simple, accurate and safe and can be used for the determination of clinical doxofylline plasma concentration.
