目的:研究结核分枝杆菌MPT64重组母牛分枝杆菌疫苗免疫学特性。方法:用分泌表达MPT64的重组母牛分枝杆菌疫苗免疫BALB/c小鼠,ELISA法检测免疫小鼠的特异性抗体滴度和抗体亚类。分离免疫小鼠脾淋巴细胞,检测淋巴细胞增殖、IFN-γ和IL-12产生水平、CD4+细胞和CD8+细胞数、脾淋巴细胞特异性CTL杀伤效应。毒株攻击后对脾脏细菌负荷计数。结果:MPT64重组母牛分枝杆菌疫苗免疫可诱导小鼠高水平的体液免疫应答,免疫小鼠脾淋巴增殖明显,IFN-γ和IL-12含量增加,CD4+和CD8+细胞百分比明显增加,CTL杀伤效应明显,对MTBH37Rv攻击后有一定的保护作用。结论:MPT64重组母牛分枝杆菌疫苗可诱导小鼠有效的体液和细胞免疫应答,有可能作为新型TB疫苗候选。
Objective: To study the immunological characteristics of Mycobacterium tuberculosis MPT64 recombinant Mycobacterium vaccae vaccine. Methods: BALB / c mice were immunized with recombinant Mycobacterium vaccae vaccine secreting MPT64, and the specific antibody titers and antibody subgroups of the immunized mice were detected by ELISA. Splenic lymphocytes were isolated from splenic lymphocytes, and the effects of lymphocyte proliferation, IFN-γ and IL-12 production, CD4 + cells and CD8 + cells and splenic lymphocyte-specific CTLs killing effect were detected. Strain bacterial load on the spleen after challenge. Results: Immunization with MPT64 recombinant Mycobacterium vaccae vaccine induced high humoral immune responses in mice. The splenic lymphocytes of immunized mice were significantly proliferated, the contents of IFN-γ and IL-12 were increased, the percentage of CD4 + and CD8 + cells was significantly increased, and the cytotoxicity of CTL The effect is obvious, there is a certain protective effect after MTBH37Rv attack. CONCLUSION: MPT64 recombinant Mycobacterium vaccae vaccine can induce effective humoral and cellular immune responses in mice and may serve as a candidate for novel TB vaccine.
