目的建立一种测定喜树叶中槲皮素和山柰素含量的高效液相色谱法。方法喜树叶样品经5%盐酸甲醇溶液水解处理后对其进行高效液相色谱法测定。色谱柱:Hubble C_(18)(4.6 mm×250 mm,5μm),流动相:乙腈-0.2%磷酸溶液(30∶70),流速:1 mL·min~(-1),柱温:30℃,检测波长:370 nm。考察该方法的专属性、标准曲线和定量下限、精密度与回收率、稳定性以及重现性。结果槲皮素和山柰素分别在5.53~69.08μg·mL~(-1)和3.99~49.90μg·mL~(-1)内线性关系良好,定量下限分别为4.23和7.81 ng,平均加样回收率分别为98.94%和98.29%,其日内、日间精密度RSD分别小于0.37%和1.04%。结论本方法是一种准确可靠、简便快捷的方法,适用于喜树叶药材质量控制研究。
Objective To establish a high performance liquid chromatographic method for the determination of quercetin and kaempferol in Camptotheca acuminata leaves. Methods Camptotheca acuminata leaf samples were hydrolyzed with 5% hydrochloric acid in methanol and determined by HPLC. The mobile phase was acetonitrile-0.2% phosphoric acid solution (30:70), the flow rate was 1 mL · min -1 and the column temperature was 30 ℃ , Detection wavelength: 370 nm. The method specificity, standard curve and quantitative limit, precision and recovery, stability and reproducibility were investigated. Results There was a good linear relationship between quercetin and kaempferol in the range of 5.53 ~ 69.08 μg · mL -1 and 3.99 ~ 49.90 μg · mL -1, respectively. The lower limit of quantification was 4.23 and 7.81 ng respectively. The recoveries were 98.94% and 98.29%, respectively. The intra-day and inter-day RSD were less than 0.37% and 1.04%, respectively. Conclusion This method is an accurate, reliable, simple and efficient method for the quality control of Camptotheca acuminata.
