目的:研究明党参根皮中异欧前胡素、欧前胡素、花椒毒酚、珊瑚菜内酯、5-羟基-8-甲氧基补骨脂素5种呋喃香豆素类成分的体外活性。方法:选取人肝癌细胞株SMMC-7721,HepG2,人肺癌细胞株A-549,人胃癌细胞株MKN-45,人宫颈癌细胞株Hela,人乳腺癌细胞株MCF-7,MDA-MB-2317种肿瘤细胞。以5×104个/mL的细胞密度接种于96孔板,给予不同浓度的药物后培养72 h,采用MTT法观察5种成分对各细胞活性的影响。结果:5种呋喃香豆素对7种肿瘤细胞均表现出不同程度的增殖抑制作用,IC50在0.30~17.23 mg.L-1之间,其中异欧前胡素效果最显著,IC50从0.39 mg.L-1到4.11 mg.L-1。结论:明党参根皮中5种呋喃香豆素类成分具有明显的抗肿瘤活性。
OBJECTIVE: To study the effects of five kinds of furocoumarin components in the root bark of minton, such as isoimperatorin, imperatorin, prickly ash toxin, coral lactone, and 5-hydroxy-8-methoxy psoralen In vitro activity. Methods: The human hepatoma cell line SMMC-7721, HepG2, human lung cancer cell line A-549, human gastric cancer cell line MKN-45, human cervical cancer cell line Hela, human breast cancer cell line MCF-7, MDA- Tumor cells. The cells were seeded in 96-well plates at a density of 5 × 10 4 cells / mL and cultured for 72 h after administration of different concentrations of drugs. The effects of five components on the activity of each cell were observed by MTT assay. Results: The five furanocoumarins showed different degrees of proliferation inhibition on seven kinds of tumor cells with IC50 of 0.30-17.23 mg.L-1, of which isoimperatorin had the most significant effect with IC50 from 0.39 mg .L-1 to 4.11 mg.L-1. Conclusion: The five furanocoumarins in the root bark of Ming dangshen have obvious anti-tumor activity.
