目的研究草乌甲素(BLA)对大鼠脊髓背根神经节(DRG)上电压门控钠离子通道Nav1.8的电生理作用。方法成年Wistar大鼠DRG细胞分离后,以10μmol/L的BLA灌流,采用全细胞膜片钳记录钠离子电流。结果 Nav1.8峰钠电流明显下降;Nav1.8稳态激活IV曲线和GV曲线向超极化方向偏移,稳态失活的GV曲线向超极化方向偏移,但这一现象没有统计学意义。结论 BLA对钠离子通道Nav1.8的电生理特性影响较轻微,可能并非临床抗炎镇痛的主要机制。
Objective To investigate the electrophysiological effects of abalone on voltage-gated sodium channel Nav1.8 in rat dorsal root ganglion (DRG). Methods Adult Wistar rats were treated with BLA at a concentration of 10μmol / L after isolation of DRG cells. Whole-cell patch-clamp was used to record the sodium current. Results Nav1.8 peak sodium current was significantly decreased; Nav1.8 steady-state activation of the IV curve and GV curve to hyperpolarization shift, steady-state inactivation of the GV curve shifted to hyperpolarization direction, but this phenomenon is not statistically Significance of learning. Conclusion BLA has a slight effect on the electrophysiological properties of sodium channel Nav1.8, which may not be the main mechanism of clinical anti-inflammatory analgesia.
