目的 :研究阿仑膦酸钠对人成骨样细胞增殖和分化的影响。方法 :进行OS 732人成骨样细胞生长曲线的绘制、MTT测定、形态学观察、碱性磷酸酶 (ALP)测定、ALP染色等。结果 :①d3时 ,与对照组相比 ,10 - 6 mol·L- 1 浓度组的细胞数目增加 2 2 .6 % ;而 10 - 5mol·L- 1 浓度组的细胞数目明显低于对照组。d 6时 10 - 6 ,10 - 7,10 - 8mol·L- 1 三种浓度组细胞数目比对照组分别增加 2 4 .7% ,2 4 .7%和 31.0 %。 10 - 5mol·L- 1 浓度组的细胞数目仍明显低于对照组。MTT测定得到了类似的结果。②加药后d4 ,10 - 6 ,10 - 7,10 - 8mol·L- 1 三种浓度组的细胞与空白对照组相比细胞数目多、形态大而规则 ,染色质深而致密 ,核质比小。③ALP染色结果显示 ,10 - 6 ,10 - 7mol·L- 1 浓度组细胞ALP颗粒染色明显深于对照组 ,并且ALP颗粒在胞质中分布均匀。ALP的定量测定结果表明 ,10 - 6 ,10 - 8mol·L- 1 浓度组细胞ALP水平明显高于对照组。结论 :阿仑膦酸钠对体外培养的OS 732人成骨样细胞具有促增殖和分化作用 ,并且这种作用具有最适合的浓度。
Objective: To study the effect of alendronate on the proliferation and differentiation of human osteoblast-like cells. Methods: The osteoblast-like growth curve of OS-732 cells was drawn, MTT assay, morphological observation, alkaline phosphatase (ALP) assay and ALP staining. Results: ① The number of cells in 10-6 mol·L-1 group increased by 22.6% at day 3, while the cell number in 10-5 mol·L-1 group was significantly lower than that in control group. Compared with the control group, the number of cells in 10, 6, 10 - 7 and 10 - 8 mol·L -1 groups increased by 24.7%, 24.7% and 31.0%, respectively. The number of cells in 10 - 5mol·L-1 group was still significantly lower than that in control group. The MTT assay yielded similar results. ② Compared with blank control group, the numbers of cells in d4, 10 - 6, 10 - 7 and 10 - 8mol·L - 1 dosing group were larger than those in blank control group, with large and regular morphology, deep and dense chromatin, Than small. ③ The results of ALP staining showed that ALP particles stained darker than control group in the concentrations of 10 - 6, 10 - 7mol·L - 1, ALP particles distributed uniformly in the cytoplasm. The results of quantitative analysis of ALP showed that ALP levels in 10 - 6, 10 - 8 mol·L - 1 groups were significantly higher than those in control group. CONCLUSION: Alendronate promotes proliferation and differentiation of OS-732 human osteoblast-like cells cultured in vitro, and this effect has the most suitable concentration.
