目的研究偏食高碘对大鼠下腔静脉肝段内皮细胞活化的影响。方法将50只断乳后约1个月的远交封闭群SPF/VAF级Wistar大鼠随机分为5组,分别为正常对照(NC)组[常规饲料(碘含量为300μg/kg),饮自来水(碘含量为5μg/L)]、偏食对照(MC)组[偏食饲料(碘含量为300μg/kg),饮自来水]、偏食+10倍碘(M10HI)组(偏食饲料,饮1 850μg/L碘溶液)、偏食+50倍碘(M50HI)组(偏食饲料,饮10 050μg/L碘溶液)、偏食+100倍碘(M100HI)组(偏食饲料,饮20 300μg/L碘溶液),每组10只,雌雄各半。采用自由饮水、摄食的方式进行染毒,连续染毒6个月。采用固相夹心酶联免疫吸附法(ELISA)测定大鼠血浆中细胞间黏附因子-1(ICAM-1)、血管性血友病因子(vWF)及血管性血友病因子前肽(vWFpp)含量。结果与正常对照组相比,无论雄性大鼠还是雌性大鼠,M50HI组和M100HI组大鼠血浆ICAM-1含量及M10HI组、M50HI组和M100HI组大鼠血浆vWF含量以及M50HI组大鼠血浆vWFpp含量均升高,差异有统计学意义(P<0.05,P<0.01);且随着摄入碘含量的逐渐升高,大鼠血浆ICAM-1、vWF含量均呈逐渐上升的趋势。结论高碘可导致偏食大鼠内皮细胞活化,且偏食引起的营养不良可加重高碘所导致的下腔静脉内皮细胞的活化。
Objective To study the effect of partial ejaculation of iodine on the activation of endothelial cells in the hepatic segment of rat inferior vena cava. The levels of intercellular adhesion molecule-1 (ICAM-1), von Willebrand factor (vWF) and vWFpp in plasma were determined by solid-phase sandwich enzyme-linked immunosorbent assay (ELISA) content. Results Compared with the normal control group, the contents of ICAM-1 in plasma of rats in M50HI group and M100HI group and those in M10HI group, M50HI group and M100HI group, and the vWFpp of M50HI group (P <0.05, P <0.01). The ICAM-1 and vWF contents in plasma gradually increased with the gradual increase of iodine content. Conclusion High iodine can lead to the activation of endothelial cells in the partial eclipse rats, and the partial eclipse-induced malnutrition may aggravate the activation of inferior vena cava endothelial cells caused by high iodine.
