目的:建立测定马钱子总生物碱复合磷脂脂质体包封率的方法。方法:采用浸渍法提取马钱子总生物碱并进行了纯化,采用硫酸铵梯度法制备了马钱子总生物碱复合磷脂脂质体。以士的宁为对照品,采用紫外分光光度法在254nm测定用破膜剂溶解的马钱子总生物碱脂质体的吸光度测定其含量。采用葡聚糖凝胶柱分离脂质体和游离药物,分别测定含量后计算包封率。结果:马钱子总生物碱平均得率为3.49%。士的宁标准曲线的范围是2.4~20μg/mL,加样回收率98.04%±1.31%。葡聚糖凝胶柱层析的回收率为102.93%±2.04%。结论:所建立的方法简便快捷稳定,可以满足马钱子总生物碱复合磷脂脂质体质量控制的需要。
Objective: To establish a method for determination of entrapment efficiency of total alkaloid phospholipid liposome. Methods: The total alkaloids were extracted by impregnation and purified. The total alkaloid phospholipid liposomes were prepared by ammonium sulfate gradient method. With strychnine as the reference substance, the absorbance of the total alkaloid liposome dissolved in the film was determined by ultraviolet spectrophotometry at 254 nm. The liposomes and free drug were separated on a Sephadex column, and the encapsulation efficiency was calculated after determining the content. Results: The average yield of total alkaloids in Strychnos nudicium was 3.49%. The calibration curve of strychnine ranged from 2.4 to 20 μg/mL, and the recovery rate was 98.04% ± 1.31%. The recovery by Sephadex gel column chromatography was 102.93% ± 2.04%. Conclusion: The established method is simple, rapid, stable and can meet the quality control requirements of total alkaloid phospholipid liposomes.
