目的 :探讨鱼精蛋白(PRM1)及锌带蛋白(ZNRD1)在结肠癌中的表达及在结肠癌生长侵袭转移中的作用。方法 :RT-PCR检测PRM1及ZNRD1在结肠癌患者和正常人中的表达;ZNRD1过表达细胞系构建及慢病毒包装PRM1-RNAi;MTT法检测细胞活力,transwell迁移和侵袭实验检测细胞迁移和侵袭能力,western blot检测Bax,Bcl-2,基质金属蛋白酶(MMP 2),MMP 9的表达。结果 :与正常组比较,结肠癌患者中PRM1表达量过高,ZNRD1表达量较低,差异具有显著性。与正常组比较,PRM1表达下降及ZNRD1表达上升能降低结肠癌细胞RKO活力和迁移侵袭能力,并下调Bcl-2,MMP 2,及MMP 9表达,上调Bax表达。结论 :PRM1及ZNRD1在结肠癌表达异常,并参与结肠癌细胞的生长侵袭转移。
Objective: To investigate the expression of protamine protamine (PRM1) and zinc banding protein (ZNRD1) in colon cancer and its role in the invasion and metastasis of colon cancer. Methods: The expression of PRM1 and ZNRD1 in colon cancer patients and normal controls were detected by RT-PCR. The ZNRD1 overexpression cell lines and PRM1-RNAi were packaged by lentivirus. The viability and transwell migration and invasion were assayed by MTT assay. Ability, western blot detection of Bax, Bcl-2, matrix metalloproteinase (MMP 2), MMP 9 expression. Results: Compared with the normal group, the expression of PRM1 in colon cancer patients was too high and the expression of ZNRD1 was low, the difference was significant. Compared with the normal group, the decrease of PRM1 expression and the increase of ZNRD1 expression could decrease the RKO activity and migration and invasion ability, and down-regulate the expressions of Bcl-2, MMP 2 and MMP 9, and up-regulate the expression of Bax in colon cancer cells. Conclusion: PRM1 and ZNRD1 are abnormally expressed in colon cancer and are involved in the growth, invasion and metastasis of colon cancer cells.
