目的:探讨针对HER2分子的RNA干涉对乳腺癌细胞生长的影响.方法:人工合成DNA,经退火磷酸化后克隆入pSUPER载体.基因转染后通过观察细胞形态和细胞计数等方法验证发夹状小干涉RNA(siRNA)对细胞生长状态的影响,以激光共聚焦检测基因表达情况.结果:siRNA表达载体转染后可以引起SKBr-3细胞大量死亡,间接免疫荧光检测出HER2蛋白表达降低.结论:针对HER2进行RNA干涉可在体外抑制SKBr-3细胞生长.
Objective: To investigate the effect of RNA interference targeting HER2 on the growth of breast cancer cells.Methods: Synthesized DNA was annealed and phosphorylated and then cloned into pSUPER vector.The expression of hairpin was verified by observing cell morphology and cell counting after transfection Small interfering RNA (siRNA) on cell growth was detected by confocal laser scanning confocal microscope.Results: Transfection of siRNA expression vector could cause a large number of SKBr-3 cell death, and indirect immunofluorescence detected HER2 protein expression decreased.Conclusion : RNA interference against HER2 inhibits SKBr-3 cell growth in vitro.
