PT-9k/129G是百日咳毒素(PT)的一种无毒性定点诱变(S1亚基中的关键氨基酸被置换)突变体。它丧失了腺苷二磷酸(ADP)核糖基转移酶活性而保留了全毒素的四级结构及真核细胞结合等其他特性。PT-9K/129G已获准作为新型百日咳无细胞菌苗的一种组分。本文通过小鼠模型评价天然或甲醛处理的PT-9K/129G的粘膜及全身免疫原性。
PT-9k / 129G is a nontoxic site-directed mutagenesis of pertussis toxin (PT) (a key amino acid substitution in the S1 subunit) mutants. It loses adenosine diphosphate (ADP) ribosyltransferase activity and retains the quaternary structure of holotoxin and eukaryotic cell binding and other features. PT-9K / 129G has been approved as a component of the new B. pertussis cell-free vaccine. The mucosal and systemic immunogenicity of native or formaldehyde treated PT-9K / 129G was evaluated by a mouse model.
